本发明制备出一种用于乳腺癌靶向治疗的鼠源性单链抗体(M4G3-scFv)并确定其DNA序列,属于生物技术领域。此单链抗体源自于鼠源性乳腺癌特异性单抗M4G3,其编码的DNA序列全长为741bp,包含编码M4G3单抗的重链可变区DNA序列375bp,以及编码M4G3单抗的K轻链可变区DNA序列321bp。本发明保留了原有抗体对于乳腺癌的特异性,又降低了对人体的抗原性,成为乳腺癌治疗的靶向载体。

The present invention is one kind of mouse originated single strand antibody, M4G3-scFv, for targeting treatment of breast cancer and its DNA sequence, and belongs to the field of biotechnology. The single strand antibody M4G3-scFv is originated specific mouse breast cancer monoclonal antibody M4G3, has a coding DNA sequence of whole length 741 bp and including one monoclonal antibody M4G3 codingvariable heavy chain region DNA sequence of 375 bp and one monoclonal antibody M4G3 coding variable light chain region DNA sequence of 321 bp. The single strand antibody M4G3-scFv has the breast cancer specificity of the monoclonal antibody M4G3 retained and lowered human antigenicity, and is used as targeting carrier for treating breast cancer.
