The present invention discloses a determination method of rate of ferrous oxidase. The method is as below: incubating a serum sample in a known amount of ferrous ions acetate buffer (0.45mol / L, pH of 5.8); oxidizing the ferrous ions into ferric ions under the effect of the oxidase; incubating for a certain period of time, adding a chromogen ferrozine to form a colored complex; measuring the content of unoxidized ferrous ions at a wavelength of 510m; and determining the concentration of ferrous oxidase by using the difference between the concentrations of ferrous ions before and after the enzymatic reaction to represent the number of oxidized ferrous ions. The determination method has short reaction time and coloration stability, meets the requirements for manual operation of an automatic biochemical analyzer and a spectrophotometer, and also improves the detection precision.
