The invention discloses a method for promoting the high-efficiency functional expression of human epidermal cell growth factor in Escherichia coli. According to the method, through the construction of expression vector pET21a-HEGF-Mxe-SUMO, high-efficiency soluble expression of hEGF protein without any exogenous amino acids in Escherichia coli BL21 (DE3) can be achieved. An effective idea and method is provided for promoting soluble expression of the exogenous protein in Escherichia coli. By fusion of a SUMO tag to the C end of a target protein, the functional expression efficiency of the fusion protein can be effectively improved, self-cleavable intein is further added into the fusion protein, and the intein and SUMO protein in the fusion protein can be cut in reducing environment so as to obtain the biological-active human epidermal cell growth factor without any exogenous amino acid sequences. The method provides a theoretical basis and new design ideas for the efficient production of complex eukaryotic biological proteins in prokaryotic expression system.
