The invention aims to provide a novel matrix formed by covalently coupling CHCA and a silicon-based mesoporous material as background interference is great during MALDI-TOF MS small molecular detection of a common matrix CHCA, wherein the background interference of the matrix is reduced. The method comprises the following steps: by virtue of an active ester method for a reaction, firstly modifying the surface of the silicon-based mesoporous material with amino; then activating CHCA carboxyl with EDC to form an amide bond; replacing EDC activated ester by NHS to form an NHS activated ester; and then combining the NHS activated ester with amino on the surface of the silicon-based mesoporous material to couple the CHCA and the silicon-based mesoporous material.
