摘要： A method of mass spectrometry or ion mobility spectrometry is disclosed in which analyte ions of a desired charge state are isolated. The method comprises: separating analytes according to their electrophoretic mobility; ionising the analytes; and mass filtering the resulting analyte ions, wherein the mass to charge ratios of the ions transmitted by a mass filter are varied as a function of the electrophoretic mobility and according to a predetermined relationship such that substantially only ions having said desired charge state are transmitted by the mass filter.

摘要： Systems and methods are provided for enabling or disabling a DMS device during an IDA method. Each cycle of the method includes a survey scan step, producing a precursor ion mass spectrum, and an MS/MS scan step. During the MS/MS scan step, one or more user-defined MS/MS scans are performed that correspond to compounds of interest found in the precursor ion mass spectrum. With each user-defined MS/MS scan, a DMS enable parameter that determines if the DMS device is enabled or disabled and one or more DMS parameters necessary to enable the DMS device for the corresponding compound of interest are received. During the MS/MS scan step, for each user-defined MS/MS scan, the DMS device is enabled or disabled according to the DMS enable parameter and one or more parameters of the DMS device are set according to the one or more DMS parameters.

The methods generally involve ionizing one or more CAH panel analytes in a sample and quantifying the generated ions to determine the amount of one or more CAH panel analytes in the sample. In methods where amounts of multiple CAH panel analytes are detected, the amounts of multiple analytes are detected in the same sample injection.

摘要： A metabolized product ion spectrum is produced for a metabolized version of a known compound using tandem mass spectrometry. Metabolized structures are inferred from the metabolized product ion spectrum. An unmetabolized product ion spectrum is received for an unmetabolized version of the known compound and unmetabolized structures are inferred from the unmetabolized product ion spectrum. Each of the metabolized structures is compared to the unmetabolized structures, producing matched and unmatched structures. For each unmatched structure, a biotransformation repository is searched for modifications and each unmatched structure and the modifications found are again compared to the unmetabolized structures, producing modified matched structures. For each atomic index of the known compound, an unmodified specificity is calculated from the matched structures, a modified intensity specificity is calculated from the modified matched structures, and a score is calculated from the specificities. Atomic indices with the highest score are identified as sites of modification.

摘要： An analytical instrument for cellular analysis of cellular particles tagged with elemental tags, such as lanthanide-based elemental tags. The analytical instrument has a sample introduction system for generating a stream of particles from the sample. An inductively coupled plasma ionization system atomizes and ionizes particles in the stream as they are received. The instrument has an ion pretreatment system and a mass analyzer. The ion pretreatment system is adapted to transport ions generated by the ionization system to the mass analyzer. The ion pretreatment system can filter out low mass ions, such as using a high-pass mass filter or a bandpass mass filter, to allow the elemental tags to pass therethrough. The mass analyzer is adapted to measure the amount of at least one element in individual particles from the stream by performing mass analysis on the ions from the atomized particles. The instrument can be adapted to measure the amount of many different tags, for example at least five different tags, at the same time to facilitate multi-parametric analysis of cells and other particles.

摘要： A method of analyzing ions is disclosed comprising performing an initial multi-dimensional survey scan comprising separating parent ions according to a first physico-chemical property and separating the parent ions according to a second physico-chemical property. Then, during a subsequent single cycle of separation, the method further comprises separating parent ions according to the first physico-chemical property. If the parent ions have a value or range of the first physico-chemical property which corresponds with the first value or range, then the method further comprises selecting parent ions of interest having a value or range of the first physico-chemical property which corresponds with the first value or range and which also have a value or range of the second physico-chemical property which corresponds with the second value or range. If the parent ions have a value or range of the first physico-chemical property which does not correspond with the first value or range then the method further comprises separating the parent ions according to the first physico-chemical property and separating the parent ions according to the second physico-chemical property.

摘要： A method of generating an inclusion list for targeted mass spectrometric analysis is disclosed. Experimentally-acquired data for a plurality of isobarically-labeled peptides of interest is received at a computer. The data includes, for each of the isobarically-labeled peptides, a mass-to-charge (m/z) ratio, a charge state, and a chromatographic retention time. Predicted properties for a corresponding unlabeled peptide are determined via the computer. The predicted properties include a predicted m/z, a predicted charge state, and a predicted chromatographic retention time. The predicted properties for each corresponding unlabeled peptide are stored to the inclusion list. The predicted chromatographic retention time for the corresponding unlabeled peptide is determined based on hydrophobicity indices of the isobarically-labeled peptide and the corresponding unlabeled peptide, and chromatographic conditions for the targeted mass spectrometric analysis.
