    • PURPOSE: A cloned animal, especially a clone calf and a preparation method thereof are provided which is capable of a high level of an organ or a medicine produced by the clone animal. CONSTITUTION: The preparation method of the cloned calf comprises the steps of : (a) obtaining and culturing the immature oocyte from an ovary of the slaughtered cattle; (b) preserving a cell line isolated from the variety of tissues; (c) removing 10-30 % of the a cytoplasm by cutting part of the oolemma of the mature oocyte of the step (a) to enucleate the oocyte; (d) injecting the donor cell line of the step (b) into the enucleated oocyte of the step (c); (e) electrofusing the designed oocyte from the step (d) at a voltage under DC condition; (f) activating the fused cell in a medium containing chemical; (g) culturing in a postactivation medium and a micro-droplet medium the activated cell and assaying the oocyte.

    • The present invention relates to a method of identifying candidate genes and the proteins encloded by them that are useful in the inducement of hybrid vigour, hybrid debility and/or the diagnosis, prognosis and treatment of disease. In particular, the present invention relates to a method for identifying candidate genes capable of producing hybrid vigour or hybrid debility in an animal or plant, comprising the steps of: (i) comparing the mRNA sequence of alleles of candidate genes isolated from an animal or plant which exhibits hybrid vigour or hybrid debility with the nucleotide sequences from the corresponding alleles isolated from the parents of said animal or plant; (ii) identifying mRNA sequence differences in the alleles from said animal or plant which exhibits hybrid vigour or hybrid debility which codes for amino acid sequence variation; and (iii) identifying that the amino acid sequence variation between alleles of the candidate gene in said animal or plant is encoded by mRNA sequences which are located within two or more different exons within the candidate gene.

    • PURPOSE: Transgenic cloned cow producing human erythropoietin and a method for producing the same transgenic cloned cow are provided, which method allows economic and effective production of various biomedicines, and facilitates production of human erythropoietin in cow's milk. CONSTITUTION: The nuclear transfer embryo produced by a fusion of a cow-derived enucleated oocyte with a nucleus of a cow-derived somatic cell transfected with a gene encoding human erythropoietin is provided, wherein the cow is an adult; the nuclear transfer embryo is SNU-B4(KCTC 10450BP). The method for producing a transgenic cloned cow expressing human erythropoietin in the mammary gland comprises the steps of: (a) introducing a gene encoding human erythropoietin into a cow-derived somatic cell line to prepare a nuclear donor cell; (b) removing cumulus cells surrounding a recipient oocyte and then the cytoplasm including the first polar body from the oocyte to prepare a cow-derived enucleated recipient oocyte; (c) transferring the transfected nuclear donor cell into the enucleated recipient oocyte and carrying out cell fusion to generate nuclear transfer embryos; and (d) transplanting the nuclear transfer embryo into a surrogate mother cow to produce live offspring.
