摘要： A molecule identification and quantitation method is provided wherein a mass tag is conjugated to an analyte and the signature ion of the mass tag remains attached to the analyte after tandem mass spectrometry fragmentation (MS-MS or MS2). Rather than losing the signature ion, a mass-balance part of the structure can be lost as a charge neutral group under tandem mass spectrometry fragmentation. The signature ion can be used for quantitation and, upon further fragmentation, can also provide ion-signals characteristic of the analyte and useful in identifying the analyte. In some embodiments, the ion-signals generated from a third mass spectrometry fragmentation (MS3) can be compared with a known mass spectrum, for example, from a look-up table, from a library, or from a database, to provide an unambiguous identification of the analyte.

The apparatus comprises at least one rectifying diode circuit for rectifying the RF voltage of the quadrupole to produce a rectified RF voltage. The apparatus further comprises at least one operational amplifier configured as a current to voltage converter, a negative input of the at least one operational amplifier connected to the output of at least one diode in the at least one rectifying diode circuit, a positive input of the at least one operational amplifier at ground, and an output of the at least one operational amplifier in a feedback look with the negative input, to reduce reverse leakage current from the at least one diode.

In one example, the method can include creating a nucleic acid ladder using CE-LIF, releasing the genome from within an adeno-associated virus, purifying said genome and analyzing said genome using CE-LIF and comparing the results of the analysis of the genome to the nucleic acid ladder to determine a size of nucleic acids in the genome.

摘要： Systems and methods are provided for microbial identification using cleavable tags. Control information is sent to a mass spectrometer to fragment one or more nucleic acid primers labeled with a first tag and monitor for an intensity of the first tag in a mass spectrometry (MS) method. An ion source provides a beam of ions from a polymerase chain reaction amplified sample that includes one or more nucleic acid primers labeled with the first tag. The first tag binds to one or more nucleic acid primers of a known microbe and is cleaved from the nucleic acid primers during the MS method. The mass spectrometer receives the beam of ions and is adapted to perform the MS method on the beam of ions. If the intensity of the first tag received from the mass spectrometer exceeds a threshold value, the known microbe is identified in the sample.
