摘要： A microfluidic device for electroelution with sample collection decoupled from the electrophoretic field can generally comprise a channel having a first fluid pathway in fluid communication with a second fluid pathway, the first fluid pathway can comprise a first port in fluid communication with a second port, and a receptacle intermediate the ports, the second fluid pathway can comprise an inlet in fluid communication with an outlet, the first and second ports can be associated with first and second electrodes, respectively, such that the electrodes can create an electrophoretic field across the receptacle, and the channel can be configured to create a pressure drop from the first fluid pathway towards the second fluid pathway that encourages the electroeluted sample to flow towards the second fluid pathway.

摘要： The invention provides a microfluidic system and method to rapidly analyze large numbers of compounds or complex mixtures of compounds, particularly, low abundance cellular proteins involved in cell signaling pathways. In one aspect, an integrated microfluidic system comprises an upstream separation module (preferably, a multi-dimensional separation device), a microfluidic device for on-device protein digestion of substantially separated proteins received from the upstream separation module, a downstream separation module for separating digestion products of said proteins, a peptide analysis module and a processor for determining the amino acid sequence of said proteins. Preferably, the system comprises an interfacing microfluidic device between the downstream separation module and the peptide analysis module.

摘要： A method of differentiating benign melanocytic nevi from malignant melanomas is disclosed. The method generally includes subjecting a skin lesion sample from a patient to mass spectrometry to obtain a mass spectrometry protein profile. This profile is compared to mass spectrometry protein profiles of reference samples, which include known normal, benign melanocytic nevi, and/or malignant melanomas. Classification of the skin lesion sample as a benign melanocytic nevus or a malignant melanoma is based on similarities and difference between the mass spectrometry protein profiles.
