摘要： Embodiments described herein generally provide for the expansion of cells in a cell expansion system using an active promotion of a coating agent(s) to a cell growth surface. A coating agent may be applied to a surface, such as the cell growth surface of a hollow fiber, by controlling the movement of a fluid in which a coating agent is suspended. Using ultrafiltration, the fluid may be pushed through the pores of a hollow fiber from a first side, e.g., an intracapillary (IC) side, of the hollow fiber to a second side, e.g., an extracapillary (EC) side, while the coating agent is actively promoted to the surface of the hollow fiber. In so doing, the coating agent may be hydrostatically deposited onto a wall, e.g., inner wall, of the hollow fiber.

摘要： The present invention relates to a hollow microfiber comprising (1) one or more cell-adhesive layers having a cell-adhesive hydrogel, (2) an outer shell layer having a high-strength hydrogel that covers the outer periphery of the cell-adhesive layer that is positioned farthest from the center axis among the one or more cell-adhesive layers, and (3) a cell layer that covers the inner periphery of the cell-adhesive layer that is positioned closest to the center axis among the one or more cell-adhesive layers. The present invention also relates to a method of manufacturing the hollow microfiber and a kit for carrying out the manufacturing method.

摘要： Embodiments described herein generally relate to systems and methods for promoting the expansion of high density non-adherent cells through the use of a cell growth chamber, a mass transfer device, and a fluid circulation loop. Improved cell growth is achieved in the cell growth chamber by using a chamber having a particular orientation and shape, e.g., conical, to create a media-rich reservoir for growing cells. By placing the chamber in a vertical position, the force of media flow along the chamber walls is substantially equal and opposite to the gravitational force on the cells. The interaction of these forces maintains the non-adherent cells in suspension. The use of the cell growth chamber in conjunction with the mass transfer device and fluid circulation loop(s) creates efficiencies by relying on the cumulative and combined features of the devices.

摘要： Initiation of growth and cultivation of cells can be performed by introducing the cells into culture compartments of a liquid-gas-phase exposure bioreactor containing a supply chamber in which there are disposed hollow-filament membranes having an inside diameter of no larger than 5 mm, wherein an inner volume of said hollow-filament membranes forms the culture compartments. Approximately one half of the supply chamber is filled with a nutrient medium and a remainder is filled with a gas mixture. Perfusion of medium and gas is turned on simultaneously or separately. The hollow-filament membranes and the cells contained therein are cyclically exposed to the gas or liquid phase.

摘要： Embodiments described herein generally relate to methods and systems for using an air removal chamber as a control for a process in a cell expansion system. The air removal chamber may be mounted on a fluid conveyance assembly for use with the system. Fluid is pumped into a fluid containment chamber of the air removal chamber, in which the level of fluid in the fluid containment chamber may be monitored through the use of one or more sensors. The sensors are capable of detecting air, a lack of fluid, fluid, and/or a gas/fluid interface, e.g., an air/fluid interface, at measuring positions within the air removal chamber. Protocols for use with the system may include one or more stop conditions. In an embodiment, the stopping of a process is automated based on the detection of air, a lack of fluid, and/or a gas/fluid interface in the air removal chamber.
