    • An improved rapid diagnostic device, assay and multifunctional buffer reagent are provided for the detection of a target analyte in a fluid test sample. The 2-step assay utilizes a dual component flow-through device comprising a test unit and a dried indicator reagent delivery unit capable of receiving the fluid sample and multifunctional buffer, respectively. The test unit comprises a reaction zone containing immobilized capture reagent that can specifically bind to the target analyte, an absorbent zone supporting the reaction zone, and optionally, a blood separation zone in lateral fluid communication with the reaction zone. The dried indicator reagent delivery unit comprises a label zone permeated with a dried indicator reagent which is capable of being placed in transient fluid communication with the reaction zone of the test unit during the assay procedure. The rapid diagnostic assay system reduces the number of assay reagents, method steps and time required for performance compared to other conventional assays.

    • The invention provides a novel array method for nucleic acid sequence detection with improved specificity which allows for detection of genetic variation, from simple SNPs (where the variation occurs at a fixed position and is of limited allelic number) to more complex sequence variation patterns (such as with multigene families or multiple genetic strains of an organism where the sequence variation between the individual members is neither fixed nor consistent). The array is comprised of short, synthetic oligonucleotide probes attached to a solid surface which are hybridized to single-stranded targets. Single stranded targets can be produced using a method that employs primers modified on the 5′ end to prohibit degradation by a 5′-exonuclease that is introduced to degrade the unprotected strand. The invention further provides for printing buffers/solutions for the immobilization of oligonucleotide probes to an array surface. The invention also provides hybridization and wash buffers and conditions to maximize hybridization specificity and signal intensity, and reduce hybridization times.

    • Embodiments of a composition for stabilizing fluorescent signal of nanoparticles and methods for its use are disclosed. In some embodiments, the composition has a pH from 7 to 10 and includes borate, protein and/or protein hydrolysate, an amine, a preservative, and a nonionic surfactant. In particular embodiments, the amine is an N-ethanol substituted amine, such as ethanolamine, diethanolamine, triethanolamine, N-methyldiethanolamine, N,N-dimethylethanolamine, or a combination thereof. In some embodiments, a fluorescent particle solution, such as a quantum dot solution or quantum dot conjugate solution, is diluted in the composition and stored at 4° C. In certain embodiments, the fluorescence intensity of the diluted fluorescent particle remains substantially the same when stored at 4° C. for at least one month or at least three months. In particular embodiments, a diluted quantum dot conjugate is used to detect a hybridized probe or a protein antigen.
