    • Acid or alternately base species concentration in aqueous and non-aqueous samples is quantitated, in the presence of interfering ionic salts, using most preferably differential conductivity detection combined with ion-exchange derivatization, wherein characteristically the acid or base species is derivatized to water, and blended indistinguishably in a required aqueous carrier phase, and the interfering salt(s) is derivatized to a distinguishable hydroxide or acid derivative(s). Using the preferred mode of detector read-out, the calibrated peak of the conductimetric response of the salt derivative(s) is compared to the peak of the conductimetric response of the sample and the response differential is displayed and used to predict the acid or base species concentration. The salt(s) concentration (with some exceptions) may be simultaneously predicted since it is proportional to the first mentioned peak. The technique has utility with respect to acids and bases broadly, and most advantageously is used with respect to strong and moderately strong acids and bases of a pK.sub.a and pK.sub.b value, respectively, of about 2 or less.

    • A simple, rapid anion-exchange column chromatographic technique is used to determine the presence of fractions of proteins and enzymes in human serum by separating the fractions on the basis of their net electrical charges. Samples of serum of unaltered ionic strength, layered on mini columns of an ion exchange agent previously equilibrated to a low ionic strength similar to blood serum with a buffer solution, are eluted stepwise with an appropriate buffer solution having different ionic strengths at each step. Elution of the samples is without previous dialysis or other ionization treatment. Column effluents are assayed for protein content or enzyme activity by conventional means, e.g., spectrophotometric analysis. Evaluation of sera from patients utilizing these methods reveals protein fractions or isoenzyme patterns which identify specific diseases associated with the protein or isoenzyme pattern.

    • A chromatographic system including cassettes, each containing a separating column which is connected at each of its ends to a coupling piece having both of its ends in the form of conical surfaces on the outside of the cassettes. Thus, the separating column cassettes may be utilized as components of various relatively simple and complex chromatographic systems.
